What is Cryo-EM?

Cryo-EM is changing the game in structural biology

Cryo-electron microscopy (cryo-EM) is leading what’s become known as the Resolution Revolution. Cryo-EM “locks” a biological sample in its natural state, allowing researchers to capture, in high resolution, interactions between molecules within proteins.   read more

Current methods such as X-ray crystallography (XRD) and nuclear magnetic resonance spectrometry (NMR) fall short when employed against large, complex and flexible structures, which include many biologically important proteins.

The process of cryo-EM is elegant in its simplicity: a sample is frozen so quickly that ice crystals do not form (vitrification), a series of thousands of images is captured, and then software reconstructs these images into a high-resolution 3D model. Cryo-EM is allowing researchers to study interactions between important proteins and dynamic events, further revealing essential structure–function relationships, which are central to structural biology.

Disease pathways are composed of complex sets of interactions, involving critical features such as membrane proteins and ion channels, which are notoriously difficult to crystallize. Cryo-EM is shedding new light on the structure and function of these features, providing researchers greater insight into them as potential drug targets.   show less

The 2017 Nobel Prize in Chemistry recognized the pioneering work of three scientists whose breakthrough developments in cryo-EM helped the structural biology community propel the method into a new era. These developments now enable institutions using Thermo Scientific cryo-EM systems to routinely produce highly resolved, three-dimensional images of protein structures.
