One of the main characteristics of imaging biological materials within their vitrified context is their low intrinsic contrast. In addition, their radiation sensitivity imposes a limit on the total dose that can be applied for imaging, which results in a poor overall signal to noise ratio in the data. Hence, within the structural biology arena there is strong need to image amorphous and/or organic (biological) materials at an improved contrast while maintaining maximum resolution.

The FEI Phase Plate offers a solution that achieves a significantly improved contrast at low spatial frequencies revealing greater levels of detail, improving the detection of weak features. With an increase in contrast, more structural and morphological information can be retrieved from e.g. cryo-tomographic reconstructions. Phase-plate TEM is particularly beneficial for cryo-tomography because the increase in image contrast allows each tilt image to be recorded at a lower electron dose resulting in less damage to the specimen.

In addition, the FEI solution offers a life time of at least four months, while its embedding into the tomography data acquisition software and strong linkage with the Falcon direct electron detector camera ensures its contribution to an increased information and high quality data output.
